Marc Buyse is a Belgian biostatistician, entrepreneur, and academic. He is the founder of One2Treat, the International Drug Development Institute (IDDI), and CluePoints, and is Associate Professor of Biostatistics at Hasselt University. He previously worked at the European Organisation for Research and Treatment of Cancer (EORTC) in Brussels and the Dana–Farber Cancer Institute in Boston.[1]
Buyse's research has focused on clinical trial methodology, meta-analysis, surrogate endpoint validation, and statistical methods in oncology. He is particularly known for his work on Generalized Pairwise Comparisons (GPC), a statistical framework for comparing treatments across multiple prioritized outcomes. In 2010, Buyse formally introduced GPC as a general method for comparing groups using multiple outcomes, including binary, continuous and time-to-event outcomes.[2]
GPC has subsequently been developed and applied across clinical research. A 2023 review in the Journal of the American College of Cardiology described GPC as a way of overcoming limitations of conventional time-to-first-event composite endpoints by allowing multiple outcomes, including the severity of events, recurrent events and clinically relevant non-survival outcomes, to be incorporated into the assessment of treatment benefit.[3] The method has been applied in cardiovascular research, oncology and benefit-risk assessment, including analyses of randomized clinical trials in which efficacy and toxicity are considered jointly.[4][5]
The GPC framework provides the basis for measures of treatment effect including Net Treatment Benefit (NTB), which summarizes the balance of favorable and unfavorable outcomes across prioritized endpoints.[6] More recent applications have used GPC and NTB to assess treatment benefit according to different priorities, including patient-specific benefit-risk assessment in the CODA trial comparing antibiotics with appendectomy for acute appendicitis.[7][3]
Buyse's 2010 paper introducing GPC has received more than 200 citations in both Scopus and Web of Science, according to the Hasselt University research repository.[8]
Buyse has served as President of the International Society for Clinical Biostatistics and President of the Quetelet Society, and is a Fellow of the Society for Clinical Trials.
