Overall, the first few sentences of your lead section are very strong and provide a good synopsis, however there are a few sentences that may need to be rephrased for clarity. First of all, the sentence "The structure can be attributed to increased stability against enzymatic degradation and improved specificity and affinity as a monomer or constituent of an oligonucleotide" may need to be split into more than one sentence and re-phrased (ie: replace the "attributed to" part). Also, saying "whenever desired" in the sentence about incorporating LNA nucleotides into RNA and DNA strands is not a very professional term.

In the Synthesis section, it might be nice to add a bit more detail. Specifically, I would suggest adding a brief summary of how LNA's are synthesized as well as a few sentences about their history (who discovered LNAs and background about how LNAs were discovered).

Regarding the LNAzymes subsection, again I would suggest adding some more details. For one, you could include a short description about what DNAzymes and how LNAzymes differ from them. Also, for the first sentence of this section "For the catalytic activity of LNAzymes" I would suggest changing it instead to "Regarding the catalytic activity."

Additionally, there are a few edits that may need to be made to the Therapeutic Applications section. For one, I would suggest re-phrasing the "in which produced" part of the sentence "Additionally, LNA has been applied to fluorescence in situ hybridization (FISH) to target the human chromosome in which produced a quick signal..." to make the meaning of the sentence more clear to readers. Also, "in situ" needs to be italicized. Another edit I would suggest to this section would be to remove the comma in the sentence "LNA genotyping assays have been conducted as well, specifically." In terms of the content of this section, it might be nice to further discuss how LNAs are incorporated into the pharmaceutical Miravirsen.

Lastly, the caption of your second image mentions "β-D configuration," but the image description doesn't define this term or mention what it is. This term also is not defined within the article. Biophysical Endosymbiont (talk) 01:00, 14 October 2020 (UTC)Reply

This is a great article, with very little I would change. The first paragraph introduces the topic in a way that can be captured by a non-specialist.

For a rewrite, I would would mainly focus on restructuring the second paragraph (ON SYNTHESIS). The information is vague. Here you mentioned the "addition of different moieties...." what are these moieties? Also you could add a little bit of background like who successfully synthesized the compound for the first time.

Again if you could re-read article from the perspective of a non-specialist, there are places you could add a little bit more context to help the reader more easily appreciate the bigger picture. For instance, you could provide a brief context on what catalysis is, what CAV-21 virus does and why cleaving the CAV-21 RNA is important. "For the synthesis..." is not a great starter for a paragraph.

In the last paragraph on "Therapeutic applications", The first statement and the one on using LNA in FISH must be cited. Overall This article is great and very informative, but again, I would add a little bit of context! Garhingh (talk) 13:58, 14 October 2020 (UTC) GarhinghReply

Overall, I think this is a really good article! It is easy to follow and the introduction is pretty clear.

It would be great if there is more information about synthesis. A little more information about the first report would be really great. Also, a little more information about the protocol and mechanism of synthesis would be really helpful. I found an article about the Locked nucleic acid synthesis(but I am not sure this would be helpful or not. if you think this is irrelevant, just ignore it).

Also, for the Therapeutic application parts "It also has potential in gene therapy to increase to potency or efficiency of certain vaccines and gene delivery systems." So, anyone started gene therapy by using LNAs? any vaccine trials? What is the current stage for LNAs therapeutic? Any company or laboratory work for this? Also, what are the obstacles to using LNAs in a therapeutic way?

Lastly, I think you missed some citations such as PCR, FISH, and things. Since Wikipedia is also for people who don't have much knowledge, the citation is always helpful!

I think this is a great start. The introduction is the strongest section. To a nonexpert unfamiliar with these terms and concepts, it is helpful to use words like “locks” to describe what is happening. I encourage you to do that more often because I think that visualization is key for a reader to best grasp these complex concepts.

I think the synthesis section should be expanded. For example, when you write about how the addition of different moieties has remained a possibility etc. It would help to elaborate on these different moieties and then use transitional sentences to get to the next idea.

I think the writing in the LNAzymes section needs to be tightened up. It would also be helpful to add more details because it is hard to follow when only big concepts are mentioned. Readers would find it helpful to have more context.

The opening sentence in the therapeutic applications section is strong. This sentence, “LNA has been applied to fluorescent in situ hybridization (FISH) to target the human chromosome in which produced a quick signal response compared to DNA oligonucleotides”, grammatically does not flow well. I would break it into two sentences and take out “in which”. It is also worth considering expanding on the potential in gene therapy in so far as it pertains to vaccines. That is really interesting and something worth expanding on for the reader to drive home the point of the real world applications.

This is looking really good so far. I would include brief description of the chemical synthesis, or just expound on how they can be obtained. I would maybe include what techniques the modified nucleosides can be used in/how they can help the research community. You could also include a brief sentence or two about how other non-natural nucleotides are used.You could also go a bit more in depth on the LNAzymes, if there is more known about them.
